Allison Baccich put DePauw ahead with just over two minutes left in the first half on an assist from Becca Gaines. Wittenberg's Rachel Dunn tied the score with 2:58 remaining on Holly McHugh's assist.
Wittenberg claimed the win just 2:57 into the first overtime as Katy Barrett scored on an assist from Dunn.
DePauw outshot Wittenberg 15-7 and held a 9-4 advantage in penalty corners.
Kaitlin Thomure recorded two saves for DePauw while Sarah Umberger had six for Wittenberg.
DePauw hosts Wooster on Saturday at noon.
